ActivityAnalysisofHAOEffectofpH,Temperature,andElectronAcceptoronHAOActivityUsingcytochromecastheelectronacceptortoreplacepotassiumferricyanideinreactionmixtures,noHAOactivitywasobserved.SDSAnalysisofHAOAminoAcidSequenceAnalysisofPurifiedHAODiscussionHAOwasconsideredtobeakeyenzymeduringthenitrogenremovalprocess,whichcanoxidizeNH2OHtonitriteornitrousoxide.

ThefindingofthepresentstudyindicatedthatthestrainY16couldoxidizeNH2OHtonitritecatalyzedbyalow-temperatureHAOduringammoniumremovalatlowtemperatures.Inthepresentstudy,ahighlypureHAOwasisolatedfromanAcinetobactersp.strainY16byananion-exchangeandgel-filtrationchromatography.PartialaminoacidsequenceanalysisofpolypeptidesofthepurifiedHAOshowedthatnohomologytothereportedHAOswaspublishedindatabase.ThedifferentsizeandaminoacidsequenceoftheHAOfromthestrainY16implythatitmaybedistinctfromotherpublishedHAOs.

Inthisstudy,thepurifiedHAOshowedahighactivitywithK3Fe(CN)6aselectronacceptor

andnoactivitywithcytochromeCaselectronacceptorwasfound.TheresultsindicatedthatalittleHAOwasextractedfromthestrainY16.Aswespeculated,tworeasonsmightcausethelowerHAOproduction.Ontheonehand,thepurificationproces
